Automated methanol induction and low pH profiling in the BioLector Pro

The methylotrophic yeasts Hansenula polymorpha (H. polymorpha or Ogataea polymorpha) and Pichia pastoris (P. pastoris) have both proven to be effective host organisms for heterologous gene expression. The capability of using methanol as a carbon source allows the isolation of strong methanol inducible promoters like formate dehydrogenase (FMD) or alcohol oxidase 1 (AOX 1) promoters. This unique property has been utilized for expression of foreign proteins through linkage of the gene of interest to the gene for alcohol oxidase. Yeast organisms preferably grow under low pH cultivation conditions between pH 4 and pH 6. Up to now, the investigated low pH range has been very challenging to measure and control in small scale cultivation systems in general Here, optical pH optodes are the measurement system of choice. In this study, we integrated recently published low pH optodes into the BioLector Pro, to perform reliable non invasive pH measurements and enables pH controlled fermentations for efficient methanol induced protein production in Pichia and Hansenula strains.
